Assessment of Cytotoxicity of Dimethyl Sulfoxide in Human Hematopoietic Tumor Cell Lines
Authors
Abstract:
Background: Dimethyl Sulfoxide (DMSO) is a solvent most broadly used as a cryopreservative agent. Antitumor effects of DMSO is a recently recognized phenomenon. In this study, cytotoxic effects of DMSO on human monocytes and T leukemic cell lines has been investigated in vitro. Methods: Human leukemic T cells (Molt-4 and Jurkat) and monocytes (U937 and THP1) were cultured in complete RPMI mediums. The cells at different logarithmic growth phases were incubated with different concentrations of DMSO (0.1, 0.2, 0.5, 1, 2 and 5%). Then viability and proliferative response of leukemic cell lines was assessed by trypan blue dye exclusion (TB test) and MTT assays, respectively. Results: DMSO has a cytotoxic effect on the leukemic cells used in this study; dose and time-dependently. This cytotoxicity for all of these leukemic cells was shown at ≥ 2% concentrations of the DMSO after 24, 48 and 72 hours’ incubation time. Moreover, there was not any significant difference between DMSO cytotoxicity in these different leukemic cell lines. Conclusion: All of the used leukemic cells showed sensitivity to DMSO at ≥2% concentrations time dependently. This sensitivity significantly increased with time. DMSO might be a cytotoxic agent for leukemic cells. It might be a useful candidate in design of chemotherapeutic protocols for leukemia as well as other cancers.

Induction of apoptosis in human tumor cell lines by platelets
Introduction: It has been reported that platelets can eradicate tumor cells in vitro, although the mechanism of this effect has not been determined. The effect of platelets on the induction of apoptosis in tumor cells is largely unknown. Materials and methods: To investigate this effect, two human hematologic cell lines, K562 and Daudi, were independently faced with unstimulated and thromb...
full textEffect of Propranolol on Angiogenic Factors in Human Hematopoietic Cell Lines in vitro
Background: Beta-adrenergic blocking agents have been broadly used for treatment of many cardiovascular diseases such as arterial hypertension and ischemic heart failure. Anti-tumoral, anti-inflammatory and anti-angiogenesis effects of propranolol (a non-selective beta-adrenergic blocker) have been shown. Angiogenesis (replenish of the pre-existing vascular networks) plays a critical role in s...
full textDown-regulation of HSP40 gene family following OCT4B1 suppression in human tumor cell lines
Objective(s): The OCT4B1, as one of OCT4 variants, is expressed in cancer cell lines and tissues more than other variants and plays an important role in apoptosis and stress (heat shock protein) pathways. The present study was designed to determine the effects of OCT4B1 silencing on expressional profile of HSP40 gene family expression in three different human tumor cell lines. Materials and Met...
full textDFT study of dimers of dimethyl sulfoxide in gas phase
Density functional (DFT) calculations at M05-2x/aug-cc-pVDZ level were used to analyze the interactions between dimethyl sulfoxide (DMSO) dimers. The structures obtained have been analyzed with the Atoms in Molecules (AIMs) and Natural Bond Orbital (NBO) methodologies. Four minima were located on the potential energy surface of the dimers. Three types of interactions are observed, CH•••O, CH•••...
full textCytotoxicity evaluation of a new series of naphthalimide derivatives against human cancer cell lines
Introduction: Cancer has been known as one the main causes of the death in the world. In recent researches, discovery of effective, selective and safe medications is a priority and emergency. In recent reports, the role of lipoxygenases (LOX) has been confirmed in neoplastic diseases. Some isoenzymes such as 5, 12 and 15 have more importance in the neoplasia. According to the efficacy of naphth...
